The debut of that comedy fundraiser got a solid start last year.
"We were quite excited and ended up raising 23 million dollars," said Richard Curtis, creator of Red Nose Day.
The red nose concept started in the UK where it's generated more than a billion dollars since 1985 in annual telethons.
"We wanna give the feeling of, people being able to directly help kids in trouble," said Curtis.
Comedian Craig Ferguson is hosting the event.
Its donations will be split 50/50 among poverty-fighting charities both in and outside of the U.S.
"It makes the world more like a small town. It's a more of a community way to help the planet," said Craig Ferguson.
That need's driven home between the laughs in a series of vignettes including Jack Black's visit to the slums of uganda motivated donors last year.
"I could feel the impact just people who came up to me on the street and said, hey i saw that thing," said Black.
"I was really moved and i gave some money to the cause."
Black's among more than two dozen celebrities taking part in performances and sketches tonight.
They're hoping to deliver on the red nose motto, good laughs for a good cause.
"Red Nose Day" airs live tonight from Los Angeles, starting at 9 p.m. on Local 4.
